Abnormal subcellular distribution of GLUT4 protein in obese and insulin-treated diabetic female dogs BJMBR
Vargas,A.M.; Barros,R.P.A.; Zampieri,R.A.; Okamoto,M.M.; de Carvalho Papa,P.; Machado,U.F..
The GLUT4 transporter plays a key role in insulin-induced glucose uptake, which is impaired in insulin resistance. The objective of the present study was to investigate the tissue content and the subcellular distribution of GLUT4 protein in 4- to 12-year-old control, obese and insulin-treated diabetic mongrel female dogs (4 animals per group). The parametrial white adipose tissue was sampled and processed to obtain both plasma membrane and microsome subcellular fractions for GLUT4 analysis by Western blotting. There was no significant difference in glycemia and insulinemia between control and obese animals. Diabetic dogs showed hyperglycemia (369.9 ± 89.9 mg/dl). Compared to control, the plasma membrane GLUT4, reported per g tissue, was reduced by 55% (P...

Characterization of mesenchymal stem cells derived from equine adipose tissue Arq. Bras. Med. Vet. Zootec.
Carvalho,A.M.; Yamada,A.L.M.; Golim,M.A.; Álvarez,L.E.C.; Jorge,L.L.; Conceição,M.L.; Deffune,E.; Hussni,C.A.; Alves,A.L.G..
Stem cell therapy has shown promising results in tendinitis and osteoarthritis in equine medicine. The purpose of this work was to characterize the adipose-derived mesenchymal stem cells (AdMSCs) in horses through (1) the assessment of the capacity of progenitor cells to perform adipogenic, osteogenic and chondrogenic differentiation; and (2) flow cytometry analysis using the stemness related markers: CD44, CD90, CD105 and MHC Class II. Five mixed-breed horses, aged 2-4 years-old were used to collect adipose tissue from the base of the tail. After isolation and culture of AdMSCs, immunophenotypic characterization was performed through flow cytometry. There was a high expression of CD44, CD90 and CD105, and no expression of MHC Class II markers. The...

Circulating levels of adiponectin and extent of coronary artery disease in patients undergoing elective coronary angiography BJMBR
Souza,R.A.; Alves,C.M.R.; de Oliveira,C.S.V.; Reis,A.F.; Carvalho,A.C..
Adiponectin (APN), an adipose tissue-released adipokine with demonstrated anti-inflammatory and anti-atherogenic properties, is encoded by a gene whose polymorphisms are associated with presence of coronary artery disease (CAD). Serum APN levels are inversely related with presence and complexity of CAD. Within this context, we sought to compare levels of total APN and its high molecular weight form (HMW APN) according to clinical presentation and extent of CAD in patients undergoing elective cardiac catheterization. From March 2008 to June 2010, clinical data and blood samples for APN and HMW APN measurements were collected from 415 subjects undergoing cardiac catheterization at two tertiary centers. CAD extent was estimated by the number of coronary...

Comparison of serum leptin, glucose, total cholesterol and total protein levels in fertile and repeat breeder cows R. Bras. Zootec.
Guzel,Saime; Tanriverdi,Meltem.
In the present study we measured serum glucose, leptin, total cholesterol and total protein concentrations in repeat breeder cows and compared them with fertile cows. For this aim, 20 repeat breeder cows and 20 fertile cows were used as material. Repeat breeder cows were found to have lower levels of leptin and glucose as compared with fertile ones. No significant differences in total cholesterol and total protein levels were observed between the two groups. No significant correlation of leptin with glucose, total cholesterol and total protein was observed in fertile and repeat breeder cows. Low concentrations of glucose and leptin can have some effects on reproductive problems as repeat breeder and help to understand potential mechanisms impairing...

Duman,Selcuk; Aktan,Tahsin Murad; Cuce,Gokhan; Cihantimur,Bulent; Tokac,Mehmet; Akbulut,Habip.
The aim of adipose tissue engineering is creating autologus vascularized fat tissue to be used for practical soft tissue reconstruction in human clinic. Unfortunately, in practice, long-term results of fat transplantation are often untrustworthy and unreliable, to overcome this problem different many lipoinjection techniques developed in the last 20 years. Centrifuge is a fundamental stepin the preparation of adipose tissue. We focused on some cell markers especially MSCs markers and histological structural properties after with lipokit centrifugation and without lipokit centrifugation of adipose tissue obtained by liposuction by this new technique. Adipose tissue was taken by liposuction and separates to two portions. One of them is centrifugated by...

Rocha,A.R.; Leite,Y.K.C.; Silva,A.S.; Conde Júnior,A.M.; Costa,C.R.M.; Silva,G.C.; Bezerra,D.O.; Cavalcante,M.M.A.S.; Feitosa,M.L.T.; Argôlo Neto,N.M.; Serakides,R.; Carvalho,M.A.M..
ABSTRACT There is a growing interest in the study of unspecialized mesenchymal stem cells, for there are still some discussions about their in vitro behavior. Regenerative medicine is a science undergoing improvement which develops treatments as cell therapy using somatic stem cells. In several studies, adipose tissue is presented as a source of multipotent adult cells that has several advantages over other tissue sources. This study aimed to characterize and evaluate the tagging of mesenchymal stem cells from the agoutis adipose tissue (Dasyprocta prymonolopha), with fluorescent intracytoplasmic nanocrystals. Fibroblast cells were observed, plastic adherent, with extended self-renewal, ability to form colonies, multipotency by differentiation into three...

Boito,Bruna; Kuss,Fernando; Menezes,Luis Fernando Glasenapp de; Lisbinski,Eduardo; Paris,Micheli de; Cullmann,Jacson Rodrigo.
ABSTRACT: The objective of this research was to examine the effect of subcutaneous fat thickness (SFT) on carcass characteristics and meat quality in 24-month-old British and crossbred British steers finished in feedlot systems. It was assessed using meta-analysis performed on data that was extracted from 46 published studies and comprising a total of 2,695 experimental units. We found that SFT was influenced by and increased linearly with hot carcass weights (HCWs) and cold carcass weights (CCWs) and their yields; moreover, the bone fraction decreased whereas the muscle and fat fractions increased with higher levels of SFT. No significant differences were detected in meat quality variables. Thus, SFT appeared to have a direct influence on HCW, CCW, and...

Isolation, Culturing, Characterization and Aging of Adipose Tissue-derived Mesenchymal Stem Cells: A Brief Overview BABT
Fathi,Ezzatollah; Farahzadi,Raheleh.
ABSTRACT The aim of this review was to describe the current state-of-the-art regarding isolation, characterization and aging of adipose tissue-derived mesenchymal stem cells (ADSCs). Mesenchymal stem cells (MSCs) have recently received widespread attention because of their potential use in tissue-engineering applications. Various studies have indicated that MSCs with a fibroblast-like morphology migrate to the sites of injury and help to regenerate damaged tissue. Over the past few years, it has been recognized that fat is not only an energy supply, but also a rich source of multipotent stem cells that can be easily harvested, isolated and selected as compared with other tissues. ADSCs are particularly interesting because of their rapid proliferation and...

DIAS JUNIOR,WALTER; BAVIERA,AMANDA M.; ZANON,NEUSA M.; GALBAN,VICTOR D.; GARÓFALO,MARIA ANTONIETA R.; MACHADO,CELIO R.; BAILÃO,ELISA F.L.C.; KETTELHUT,ISIS C..
ABSTRACT Adaptive changes of carbohydrate and lipid metabolism induced by 7, 15, 30, 60, 90, 150 and 200 days of fasting were investigated in red tilapia (Oreochromis sp.). Plasma glucose, lactate and free fatty acids (FFA) levels, liver and muscle glycogen and total lipid contents and rates of FFA release from mesenteric adipose tissue (MAT) were measured. Plasma glucose levels showed significant differences only after 90 days of fasting, when glycemia was 34% lower (50±5mg.dL-1) than fed fish values (74±1mg.dL-1), remaining relatively constant until 200 days of fasting. The content of liver glycogen ("15%) in fed tilapia fell 40% in 7 days of food deprivation. In 60, 90 and 150 days of fasting, plasma FFA levels increased 49%, 64% and 90%, respectively,...

New concepts in white adipose tissue physiology BJMBR
Proença,A.R.G.; Sertié,R.A.L.; Oliveira,A.C.; Campaãa,A.B.; Caminhotto,R.O.; Chimin,P.; Lima,F.B..
Numerous studies address the physiology of adipose tissue (AT). The interest surrounding the physiology of AT is primarily the result of the epidemic outburst of obesity in various contemporary societies. Briefly, the two primary metabolic activities of white AT include lipogenesis and lipolysis. Throughout the last two decades, a new model of AT physiology has emerged. Although AT was considered to be primarily an abundant energy source, it is currently considered to be a prolific producer of biologically active substances, and, consequently, is now recognized as an endocrine organ. In addition to leptin, other biologically active substances secreted by AT, generally classified as cytokines, include adiponectin, interleukin-6, tumor necrosis factor-alpha,...

Obesity, inflammation, and insulin resistance BJPS
Martins,Luana Mota; Oliveira,Ana Raquel Soares; Cruz,Kyria Jayanne Clímaco; Torres-Leal,Francisco Leonardo; Marreiro,Dilina do Nascimento.
White adipose tissue (WAT) is considered an endocrine organ. When present in excess, WAT can influence metabolism via biologically active molecules. Following unregulated production of such molecules, adipose tissue dysfunction results, contributing to complications associated with obesity. Previous studies have implicated pro- and anti-inflammatory substances in the regulation of inflammatory response and in the development of insulin resistance. In obese individuals, pro-inflammatory molecules produced by adipose tissue contribute to the development of insulin resistance and increased risk of cardiovascular disease. On the other hand, the molecules with anti-inflammatory action, that have been associated with the improvement of insulin sensitivity, have...

Polymorphism of the BCO2 gene and the content of carotenoids, retinol, and α-tocopherol in the liver and fat of rabbits R. Bras. Zootec.
Strychalski,Janusz; Gugołek,Andrzej; Brym,Paweł; Antoszkiewicz,Zofia; Chwastowska-Siwiecka,Iwona.
ABSTRACT The study was carried out to evaluate the effect of AAT-deletion mutation at codon 248 of the BCO2 gene on the content of lutein, β-carotene, retinol, and α-tocopherol in the liver and fat of crossbred rabbits. The experimental animals comprised 90 rabbits, produced by reciprocal crossing between ins/del heterozygous parents of Flemish Giant, New Zealand Red, and Termond White breeds. All rabbits in the litter were genotyped, given the same diet, and finally slaughtered at 140 d of age. It was found that regardless of the mating scheme employed, all del/del homozygotes had the yellow fat trait, whereas heterozygous and homozygotes animals without the AAT deletion (ins/ins) developed white fat. Lutein concentration in fat was over 13-fold higher in...
